Effect of Etizolam (1.0mg) on Sleep in Patients with Chronic Insomnia: Polysomnographic Finding

Abstract
Etizolam is a new short-acting thienodiazepine which shows clinical anti-anxiety. anti-depressive. anti-panic. muscle relaxant effects as well as sedative/hypnotic effects.
The aims of this study were, to evaluate objectively the effect of etizolam 10 mg on sleep using polysomnography and to investigate daytime sleepiness and performance levels for possible residual effects of the drug.
To date. six patients (4 males and 2 females age 35~52) suffering from chronic insomnia have been studied. Subjects attended the laboratory for two consecutive nights taking placebo. These screening nights were followed by two treatment blocks
(etizolam
1.0mg and placebo) in a double-blind. counterbalanced, crossover design. Each block consisted of two consecutive days. the first being for adaptation and the second being for testing. Blocks were separated from each other by 7 day drug washourt
period
which the subjects spent at home. One night of polysomnography followed by self-assessment of drug effects on sleep. a MSLT and several performance tests (digit symbol substitution. simple reaction time. choice reaction time. pegboard test and
finger
tapping test) were conducted on the sccond day of each block.
Etizolam showed an hypnotic effect in all the patients. Compared to placebo. etizolam significantly increased total sleep time. reduced sleep latency. and decreased time awake during sleep. Etizolam did not produce daytime sleepiness and
performance
impairment the following day.
The result of this study indicates that etizolam 1.0mg can be a effective and safe treatment for chronic insomnia. To delineate the hypnotic characteristics of etizolam in detail. however. the dose-related effects of etizolam on sleep should be
investigated along the effects of repeated administration. in consequent studies.
